Epitalon (also spelled Epithalon or Epithalone) is a synthetic tetrapeptide with the amino acid sequence Ala-Glu-Asp-Gly (molecular formula C14H22N4O9). It is a derivative of Epithalamin, a polypeptide naturally produced by the pineal gland, and was originally developed by Russian scientist Vladimir Khavinson at the St. Petersburg Institute of Bioregulation and Gerontology.
The primary mechanism centers on telomerase activation. Telomeres — the protective caps at the ends of chromosomes — shorten with each cell division, eventually leading to cellular senescence and aging. By upregulating telomerase production, the compound promotes telomere renewal, which is the foundation of its anti-aging applications in modern peptide therapy.
The compound is not FDA-approved for human use. It is available as a research chemical and has been studied primarily in Russian clinical trials and animal models.
Key Takeaways
- Epitalon is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) derived from the pineal gland polypeptide Epithalamin.
- It activates telomerase, the enzyme that renews telomeres at the ends of chromosomes, slowing cellular aging.
- It also stimulates melatonin production, improving sleep quality and circadian rhythm.
- Standard Epitalon dosage: 5-10 mg/day for 2-3 weeks, repeated 1-2 times per year.
- Epitalon side effects are minimal, primarily mild nausea and injection site reactions.
What is Epitalon?

Origin and Development
The compound was developed as a synthetic version of Epithalamin, a naturally occurring polypeptide secreted by the pineal gland. The pineal gland is a small endocrine gland located deep in the brain, historically referred to as the “third eye” due to its role in regulating circadian rhythms and its sensitivity to light-dark cycles.
Vladimir Khavinson’s research at the St. Petersburg Institute of Bioregulation and Gerontology established the foundation for Epithalon peptide therapy. His work focused on bioregulators — short peptide sequences that regulate specific organ functions — and demonstrated that pineal peptides could influence aging markers in both animal and human studies.

N-Acetyl Epitalon Amidate
N-Acetyl Epitalon Amidate (also sold as N-Acetyl Epithalon Amidate) is a modified version of the same Ala-Glu-Asp-Gly sequence. An acetyl group is added at the N-terminus and the C-terminus is amidated. Both are common peptide modifications: capping the two ends removes their charges and makes the peptide harder for enzymes to break down, which is meant to give it more stability and a longer active life in the body.
Nearly all Epitalon research, including the Russian clinical work and the animal lifespan studies, used the unmodified peptide. No published head-to-head studies show that the modified form works better, or that it survives the digestive tract well enough to replace injection. Treat claims of extra potency as unproven; dosing for it is extrapolated from standard Epitalon.
How Epitalon Works
Telomerase Activation
Telomeres are repetitive nucleotide sequences that cap the ends of chromosomes, protecting genetic information during cell division. Each time a cell divides, telomeres shorten slightly. When telomeres become critically short, the cell enters senescence (stops dividing) or undergoes apoptosis (programmed death). This progressive telomere shortening is one of the primary mechanisms driving biological aging.
Telomerase is the enzyme responsible for rebuilding telomeres after cell division. Its production declines with age, which accelerates telomere shortening and the aging process. The compound upregulates telomerase synthesis by the pineal gland, promoting telomere renewal and extending the functional lifespan of cells.
Melatonin Regulation
The peptide stimulates melatonin production and regulates its secretion pattern. Melatonin is the primary hormone governing the sleep-wake cycle (circadian rhythm), and its production also declines with age — which is why sleep quality typically deteriorates in older adults. By restoring melatonin synthesis, the compound supports deeper, more restorative sleep and normalized circadian function.
Antioxidant Activation
Research suggests that the peptide activates endogenous antioxidant enzymes, including glutathione peroxidase and superoxide dismutase. These enzymes neutralize free radicals and inhibit lipid oxidation, reducing oxidative stress — a key contributor to cellular damage and age-related disease progression.
Epitalon Benefits
Anti-Aging and Longevity
The primary Epitalon benefits center on telomerase-mediated cellular rejuvenation. Animal studies have demonstrated measurable life span extension in aging rats treated with Epithalon peptide, consistent with the telomere-lengthening mechanism observed in cell culture studies. Human clinical trials conducted at the St. Petersburg Institute showed reduced markers of biological aging in treated subjects.
In female SHR mice given Epitalon from 3 months of age until natural death, average lifespan did not change, but the longest-lived 10% of mice and the maximum lifespan both rose by about 12-13% versus untreated controls. The treated mice also had about 17% fewer chromosome aberrations in bone marrow cells and less leukemia. In fruit flies (Drosophila melanogaster), Epitalon extended lifespan by 11-16%.
The anti-aging effects extend beyond lifespan. Research suggests that treatment may reduce the risk of age-related neurodegenerative conditions by maintaining cellular health in neural tissue. These findings are preliminary but align with the known relationship between telomere length and neurodegenerative disease risk.
Sleep Quality Improvement
Through its stimulation of melatonin production, the compound produces measurable improvements in sleep architecture. Users consistently report deeper, more restorative sleep during treatment cycles. For bodybuilders and athletes, improved sleep quality directly supports recovery, growth hormone secretion, and overall training performance.
The recommended timing of administration — 1-2 hours before bedtime — takes advantage of this melatonin-stimulating effect to promote natural sleep onset.
Immune System Modulation
The peptide regulates T-cell function by stimulating interferon-gamma production. T-cells are critical components of the adaptive immune system, and their function declines with age (immunosenescence). By supporting T-cell activity, this compound may help maintain immune competence in aging populations.
Skin Rejuvenation
The compound inhibits the synthesis of MMP9 protein, which plays a role in the breakdown of extracellular matrix components including collagen and elastin. By reducing MMP9 activity, it supports the structural integrity of skin tissue, potentially improving skin texture and reducing visible signs of aging.
Oxidative Stress Protection
The activation of antioxidant enzymes provides cellular protection against oxidative damage. This mechanism has generated research interest in the peptide’s potential to reduce the risk of conditions driven by oxidative stress, including cardiovascular disease and certain inflammatory conditions.
Epitalon Dosage and Protocol
Dosage Guidelines
| Parameter | Recommendation |
|---|---|
| Epitalon dosage range | 5-10 mg/day |
| Cycle duration | 2-3 weeks |
| Frequency | 1-2 cycles per year |
| Break between cycles | 4-6 months |
| Administration timing | 1-2 hours before bedtime |
Start at the lower end of the dosage range (5 mg/day) for the first cycle to assess tolerance. Higher doses can be split into two equal administrations — morning and evening — to maintain more consistent blood levels.
Administration
The compound is most commonly administered via subcutaneous injection, following standard peptide injection protocols. The lyophilized powder is reconstituted with bacteriostatic water before injection.
| Form | Bioavailability | Notes |
|---|---|---|
| Subcutaneous injection | Highest | Preferred method; rapid absorption |
| Intramuscular injection | High | Alternative injection route |
| Nasal spray | Moderate | Convenient but less predictable absorption |
| Oral capsules | Low | Significant degradation in GI tract |
Injectable administration is strongly preferred because this peptide undergoes extensive degradation in the digestive tract. Oral and nasal forms deliver significantly less active compound to the bloodstream.
Cycling Rationale
The 2-3 week cycle with 4-6 month breaks reflects the biological timeline of telomerase activation and telomere renewal. Short, intense cycles stimulate the telomerase response, and the breaks allow the body to consolidate the cellular benefits before the next cycle. Extended continuous use has not been studied and is not recommended.
Stacking
The peptide is sometimes stacked with Thymalin, a bioregulator peptide that enhances immune function through T-cell maturation. The combination provides synergistic immune and anti-aging effects. Animal studies at the St. Petersburg Institute demonstrated increased lifespan in subjects receiving both peptides compared to either alone.
It can also be used alongside GH secretagogues or recovery peptides without hormonal interference, as its mechanism is entirely independent of the hypothalamic-pituitary axis.
Epitalon Side Effects
Epitalon side effects are generally minimal. The peptide’s mechanism of action targets telomerase and melatonin pathways rather than hormonal systems, which limits the scope of potential adverse effects.
| Side Effect | Frequency | Notes |
|---|---|---|
| Nausea | Uncommon | Mild and transient; more common in first days |
| Injection site reaction | Uncommon | Redness or minor swelling at injection site |
| Drowsiness | Occasional | Related to melatonin stimulation; expected when taken before bed |
| Headache | Rare | Typically mild |
No serious adverse effects have been reported in the available clinical literature. However, the limited scope of human clinical trials — primarily conducted at a single research institute — means that the full adverse effects profile may not be completely characterized.
Theoretical Considerations
Because the compound promotes cell proliferation through telomerase activation, there is a theoretical consideration regarding its use in individuals with existing cancerous or pre-cancerous conditions. While no direct link to cancer promotion has been established — and some research suggests potential anti-tumor properties — the proliferative mechanism warrants caution in at-risk populations.
The Bottom Line
Epitalon’s telomerase-activating properties directly address one of the fundamental drivers of biological aging (telomere shortening) while its secondary effects on melatonin production, antioxidant activation, and immune modulation provide additional health-span benefits.
- Its favorable side effect profile, non-hormonal mechanism, and short cycling requirements (2-3 weeks, 1-2 times per year) make it an accessible addition to anti-aging and longevity-focused protocols.
- The primary limitation remains the relatively narrow evidence base, with most human clinical data originating from a single research institute.

Can you buy Epitalon in a pre mixed pen.
Can you take epitaph in conjunction with ipamorelin/cjc1295
Thanks, Cheri
How much bacteriostatic water would one have to inject to reconstitute 50 mg epitalon and then administer 5 mg daily? thank you
You can use 10,000 gallons of water. The more you use, the less diluted the product, the more liquid you need to inject. Use this calculator for help https://muscleandbrawn.com/peptides/how-to-reconstitute-peptide/
You mention 100mcg earlier in the article, and then refer to the Russian 5-10mg per day study. I read that epithalamin extract was used for the Russian study, and that the peptide Epitalon is not needed at the same dosage. Can you please confirm or elaborate on your dosing schedule?
Thinks it’s the difference between oral and injectables
I got some Epitalon and when I inject subq into my abdomen area it stings. Why is this?
Most injectable things sting. Unless you have a severe reaction, you’re fine
Nothing stings like Epitalon. It’s the king of sting.
Non
bonjour peut on utliser epitalon et melanotan2 ?merci
The article says the optimal dosage is 5-10 mg per day abut other protocols dosage in the micrograms if taken every day? What is the best guidance on dosage?
The article states that bodybuilders take 75mcg, whereas a study used mg doses?
I am confused about the dose because I’ve read 1.5ml, 30 units, 5mg, 10mg. So how would I know which injections to begin with. I’d prefer to start lower and move my way up of needed. One article says to do this once a year, another say do this every 6 month. Then another says ever 3 or 4 months. The information seems very conflicting and want to start off right then maintain it appropriately.
It’s misinterpretation from the Russian studies. Even in this article there’s 50x dosage conflict. Apparently 5-10mg was a wrong dosage because it was based on a completely different compound.
What is the correct dosage? There are very different recommendations on the Internet.
Curious as to how long epitalon will be safe in the refrigerator after reconstitution
Looks like many of us have the same question, so asking again in case this triggers a response.
Dosing question.
I’m wanting this for longevity and anti-aging. Not specifically for melatonin production. I sleep fine.
I don’t speak Russian, but I keep hearing from people that the original studies that were 5 to 10 mg per day.we’re based in epithalimin not epithalon/epitalon.
The former being a compound and extract and requiring a much higher dosage.
Again, I don’t speak Russian, but don’t want to be taking 50 to 100 times the recommended dose because we don’t have safety protocols in place for it yet.
But I also would prefer not to take it and have it do absolutely nothing.
Ideally, looking for a tea lengthening and overall anti aging/skin rejuvenation.
What I am reading on various forums from people who do speak Russian is that 100 µg, to 200 µg, -or up to 1 mg per day at the absolute most (NOT 5-10 mg/day) is proper dosing g for epithalon
I feel like most people are just regurgitating the same information over and over, but I can’t find any actual references to the actual studies to get to the truth.
Would love to hear, especially if you have a reference link, what the antiaging/skin benefit protocol would be
how much bacteriostatic water I have to put in a 50mg vial, and the amount of Unit daily
